Description

Principle of the assay: mouse ANGPTL3 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for ANGPTL3 has been precoated onto 96-well plates. Standards(sf21, S17-T455) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for ANGPTL3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse ANGPTL3 amount of sample captured in plate.
Background: ANGPTL3, also known as Angiopoietin-like 3 or HYPLl, is a protein that in humans is encoded by the ANGPTL3 gene. It is mapped to 1p31.3. The protein encoded by this gene is a member of the angiopoietin-like family of secreted factors. It is expressed predominantly in the liver, and has the characteristic structure of angiopoietins, consisting of a signal peptide, N-terminal coiled-coil domain, and the C-terminal fibrinogen (FBN)-like domain. ANGPTL3 also acts as dual inhibitor of lipoprotein lipase (LPL) and endothelial lipase (EL), and increases plasma triglyceride and HDL cholesterol in rodents. It can inhibit endothelial lipase to catalyze HDL-phospholipid and increase HDL-PL levels
